If the backstory of a character in their original universe involved them losing a close family member or loved one through a tragic accident, you might slightly rework that. You can tweak the characters, but it should be in ways that are consistent with their essence. If you've lost them because you tore up the blueprint for the characters, you'll be fighting an uphill battle to get anyone engrossed in your story.
